Read with verse 31. The two close the chapter as a pair.
Quick answer
No amount of cleverness wins an argument with God. The verse stacks three words for smart and cancels all of them at once.
Read both lines
Three nouns pile up, wisdom, understanding, counsel, which are the book's own highest goods and the things it has spent twenty chapters recommending. That is what makes the verse startling: it is not dismissing wisdom, it is saying where wisdom's limit is. The last word is the preposition. Against the LORD, set opposite Him, is the only position from which these become worthless, and the verse says nothing at all against wisdom used any other way. So this is not anti-intellectual. It is a statement about which direction thinking is pointed.
